Had a Tune-up done Plugs, wires,oil change,and filters all replaced now there is a knocking sound and the diagnostic says there is a misfire in the 3rd cylinder is the mechanic that did the tune-up responsible for the repairs that need to be done now?

If the mechanic provided you the parts and one of the wires, plugs is bad then yes he should be.
but if it turn out some thing else then he is not.
unless he dropped some thing in there and cause the valve or piston goes bad then he is the one to blame.
