I have worked on a few single cylinders that had the same symptoms and it was the intake rocker arm came loose and fell down and was not allowing any fuel into the cylinder

For it to run you need fire fuel and compression. you say you have fire. if you spray starting fluid in the carburetor then you should have fuel. pull the plug out and check the compression. sounds like a valve or valve seat is to blame.
