My drive will now only start in the "Manual" power mode but my computer does not see it. I dont know why this is happening. Can I still pull the data and transfer to a new drive?

However, as it is not showing up the controller in the Buffalo case is most likely dead. You will need to take it to a local computer store and purchase a new enclosure to put it in, then you should be ok.
If it doesn't work in the new enclosure then you have an actual HDD problem and may need the help of a pro. Let me know if this is the case and I'll make a recommendation based on your location(need to know location).
