If you are sure that your mic is working properly, then take a look at this illustrated picture:
On the skype menu, click "Call" and select "Audio Settings", this window will open up, the first slider (option) is for Microphone.. when you speak on the Mic, the indicator will show the status. Simply move the first slider to the extreme right and save the settings. If this doesn't seem to work, then you need to check your computer sound settings. Open the sound settings and make sure that Microphone is selected as the default recording device, and is not muted.
Since you didn't mention the details about your computer, I can't make this specific. If you need further help, let me know with more details like the Operating System installed on your computer, the sound card...etc...
