It sounds to me that you have antifreeze/coolant leaking out of the weep hole on the transmission inspection cover. If that is the case, then yes. The transmission has to be removed to replace the leaking freeze plug. However, Before having that done, check the large coolant line exiting the engine just above the transmission. There is a large "O" ring on it that may cause it to seem to be leaking between the transmission and the motor. Also, the coolant tube (same one) may have a pin hole in it.
Commonly known as a frost plug / welch plug, If its the rear one then yes the trans has to come out. make sure the garage fits a brass one as they do not corrode.
