If a building is "wheelchair accessible," it means that it is constructed to allow a person using a wheelchair to move around without being obstructed, and with access to all the features of that building including its bathrooms and multiple levels.
I'd like to add that you need to careful when inquiring if a place is wheelchair accessible. I'll use hotels as an example because I've traveled a lot and have learned about accessibility the hard way sometimes. Some hotels say a room is wheelchair accessible because, there are bars next to the toilet, but, come to find out you can't fit a wheelchair into the bathroom. Other hotels have a huge bathroom including a roll-in shower and provide some type of shower bench. So, know what your needs are and be specific about how wheelchair accessible a place is when asking.
I have learned that wheelchair accessible often does NOT mean that. It means that there is a sign that says it's wheelchair friendly. The door may weight 50 lbs and open towards you so you have to try and pull it open while wheeling yourself - which takes two hands. I could go on.. but I won't. ;)
